Description: Full length recombinant human SMARCA2 Remodeling Enzyme. SMARCA2 is a core ATPase subunit of the ATP-dependent chromatin remodeling complex SWI/SNF that mobilizes nucleosomes and plays key roles in gene transcription regulation, development, genome replication and repair [1,2]. Aberrant activity of SWI/SNF subunits is linked to more than 20% of human cancers and multiple developmental disorders, making SMARCA2 an emerging candidate therapeutic target [3,4]. SMARCA2 can be used in conjunction with EpiDyne®-FRET Nucleosome Remodeling Assay Substrate (EpiCypher 16-4201) in HTS-compatible drug discovery studies.

This product is sufficient to perform 100 remodeling reactions using EpiDyne®-FRET substrate (EpiCypher 16-4201). A single reaction is defined as 10 µL containing 15 nM SMARCA2 (concentration is lot-specific), 20 nM EpiDyne®-FRET, & 1 mM rATP with remodeling to completion in <30 minutes. Remodeling readout can be observed in the change of Cy3/Cy5 ratio over time in a black 384-well assay plate on a compatible fluorescent plate reader. 5x SMARCA Remodeling Assay Buffer is included (EpiCypher 21-0014; 100 mM Tris HCl pH 7.5, 250 mM KCl, 15 mM MgCl2, 0.05% (w/v) BSA, 0.05% (v/v) Tween 20).
