Description: Histone post-translational modifications (PTMs), such as lysine methylation and acetylation, interact with reader, writer, and eraser proteins as well as with other combinatorial PTMs, forming part of the "histone code." This code functions as a regulatory language governing chromatin structure and gene expression. Recombinant nucleosomes with combinatorial PTMs provide valuable tools for studying the biological functions of multiple PTMs and PTM crosstalk. H3K4me3,K9,14,18ac (histone H3 lysine 4 trimethylation, lysine 9, 14, and 18 acetylation) Recombinant Nucleosome, Biotinylated consists of 147 base pairs of 601 sequence DNA [1] wrapped around an octamer of core histone proteins (two each of H2A, H2B, H3.2, and H4) to form a nucleosome, the basic repeating unit of chromatin. The DNA contains a 5' biotin-TEG group. H3K4me3,K9,14,18ac nucleosome contains H3.2 with trimethylated lysine at position 4 and acetylated lysine at positions 9, 14, and 18. Histone H3.2 also contains a Cys to Ala substitution at position 110. H3K4 methylation and H3 acetylation are highly linked: hyperacetylation of the H3 N-terminal tail (e.g. H3K9,14,18ac) promotes H3K4 methylation by increasing tail accessibility for H3K4 methyl writers such as the methyltransferase MLL1 [2].

H3K4me3,K9,14,18ac nucleosome is highly purified and suitable for a variety of applications, including use as a substrate in enzyme assays, high-throughput screening and inhibitor testing, chromatin binding studies, protein-protein interaction assays, structural studies, and in effector protein binding experiments.
