Description: EpiCypher dNucs™ are semi-synthetic recombinant nucleosomes, containing one or more histone post translational modifications (PTMs). Nucleosomes, the basic repeating unit of chromatin, are the natural targets of chromatin readers and modifying enzymes, providing a physiologically relevant substrate for characterization of epigenetic proteins. dNucs consist of 147 base pairs of DNA wrapped around an octamer core of histone proteins (two each of H2A, H2B, H3.3, and H4). The 147 bp 601 sequence, identified by Lowary and Widom [1], has high affinity for histone octamers and is useful for nucleosome assembly. H3.3S31phos dNuc contains phosphoserine at position 31 on histone H3.3, which has shown to be a negative regulator of KDM4B binding, and thereby preserving H3K9me3 marks [2]. The DNA in this nucleosome contains a 5'biotin-TEG group.
