Triple IHC

GFAP stained for astrocytes. Vascular-associated astrocytes in the young adult rat retina
Tissue specimen of 3 month old (young adult) Wistar rat retina illustrates vascular-associated astrocytes in the mid-peripheral retina. Triple label immunohistochemistry was executed on retinal wholemount preparations. GFAP stained for astrocytes (red, Cy3-conjugated mouse monoclonal anti-GFAP), GS Isolectin B4 stained for blood vessels (blue, Cy5-conjugated streptavidin), and S-100 stained for cell soma region of astrocytes (green, Alexa 488-conjugated anti-rabbit IgG). Equipment used:
Carl Zeiss LSM 510 META inverted confocal microscope equipped with UV-Argon laser scanning, Axiovert 200M and LSM 510 scan head. The objective lens used was a Plan-Apochromat 40x/0.5. Alexa488 fluorescence was excited at 488 nm, Cy3 at 561 nm and Cy5 at 633 nm. Image captured of optical z-series projection, resolution of 1024 x 1024, and data depth of 12.
